اثر عصاره آبی الکلی سداب بر روی شاخص‌های استرس اکسیداتیو و آنزیم کولین استراز در بافت مغز موش‌های صحرایی دریافت‌کننده رژیم پر کلسترول

چکیده

مقدمه و هدف: بروز بیماریهای قلبی و عروقی و استرس اکسیداتیو با رژیم پر کلسترول ارتباط مستقیم دارند.گیاه سداب از گیاهان دارویی مورد استفاده در طب سنتی ایران و ملل مختلف بوده و با اثرات آنتی اکسیدانی خود در درمان بسیاری از بیماریها کاربرد داشته است. هدف از این مطالعه بررسی اثر عصاره هیدروالکلی گیاه سداب بر روی آنزیم کولین استراز  و شاخص های استرس اکسیداتیو در بافت مغز موش های دریافت کننده  رژیم پرکلسترول می باشد.  
 
 مواد و روش­ها : در این مطالعه تجربی، 28 موش صحرایی نر نژاد ویستار به طور تصادفی به 4 گروه کنترل ،کنترل ودریافت کننده سداب ، رژیم غذایی پر کلسترول   و رژیم غذایی پر کلسترول و دریافت‌کننده سداب تقسیم شدند. برای مدت 8 هفته، گروه های کنترل و کنترل و سداب، غذای معمولی و دو گروه دیگر غذای پر کلسترول (کلسترول 1% و اسیدکولیک 25/0 %) را دریافت کردند. پس از 8 هفته گروه‌‎های تحت درمان، با mg/kg100 عصاره سداب به صورت داخل صفاقی به مدت 3 هفته درمان شدند. در پایان کار، موش ها با اتر بیهوش شده و مغز آنها خارج و پس از تهیه هموژنه بافتی، فعالیت آنزیم بوتیریل کولین استراز و سنجش استرس اکسیداتیو (مقادیر گونه های فعال اکسیژن (ROS)، گلوتاتیون و فعالیت کاتالاز) با استفاده از کیت های اختصاصی انجام شد.
 
نتایج: هشت هفته رژیم پرکلسترول موجب کاهش میزان گلوتاتیون و کاهش فعالیت آنزیم های بوتیریل کولین استراز و کاتالاز و افزایش ROS در بافت هیپوکمپ موش های صحرایی گردید. درمان سه هفته‌ای با عصاره هیدروالکلی سداب موجب افزایش معنی دار فعالیت آنزیم بوتیریل کولین استراز هیپوکمپ موشهای تحت درمان در مقایسه با گروه پرچرب شد. به علاوه فعالیت کاتالاز و میزان گلوتاتیون احیا در گروه پرچرب تحت درمان در مقایسه با گروه پرچرب افزایش پیدا نمود که معنی دار نبود.همچنین  تغییرات  میزان ROS در گروههای مختلف  معنی دار نبود.
 
نتیجه­ گیری: نتایج این مطالعه نشان داد عصاره هیدروالکلی سداب موجب افزایش معنی دار فعالیت آنزیم بوتیریل کولین استراز هیپوکمپ موش های تحت درمان در مقایسه با گروه پرچرب شده است.  ضمنا  عصاره سداب سطوح آنتی اکسیدانی و دفاع در برابر استرس اکسیداتیو در موش‌های دریافت کننده رژیم پرکلسترول ر اتغییر داده  است اما این تفاوت از نظر آماری معنی‌دار نبود.

The effect of hydroalcoholic extract of Ruta graveolens on oxidative stress markers and cholinesterase in brain rats receiving high-cholesterol diet

Background and Objective: High cholesterol is associated with higher incidence of cardiovascular diseases and oxidative stress. Ruta graveolens (RG) is one of the medicinal plants used in Iranian traditional medicine and different nations in the treatment of many diseases with antioxidant effects. The aim of this study was to investigate the effect of hydroalcoholic extract of RG on oxidative stress markers and cholinesterase enzyme in brain tissue of rats receiving high-cholesterol (HC) diet.
 
Materials and Methods: In this experimental study, 28 male Wistar rats were randomly divided into 4 groups: Control, RG and HC diet and  HC diet+RG. For 8 weeks, Control and RG group received normal diet and the other 2 groups received HC diet (1% cholesterol and colic acid 0.25%). After 8 weeks, treatment groups were treated with 100 mg/kg of RG intraperitoneally for 3 weeks. At the end of the experiment, the rats were
anesthetized with diethyl ether and their brain were excised. After tissue homogenate was prepared, oxidative stress markers (level of reactive oxygen species (ROS), glutathione and catalase activity) and butyrylcholinesterase activity were measured using specific kits.
 
Results: Eight-week diet with HC decreased glutathione level and the activity of butyrylcholinesterase and catalase enzymes and increased ROS in the hippocampal tissue of rats. Three-week treatment with hydroalcoholic extract of RG significantly increased the activity of the butyrylcholinesterase enzyme in the treated rats compared with the HC diet group. In addition, the activity of catalase and level of glutathione increased in HC diet+RG group compared to the HC diet group. However, this increase was not significant. Also, the difference between level of ROS in different groups was not statistically significant.
 
Conclusion: Results of this study demonstrated that hydroalcoholic extract of RG significantly increases the activity of the butyrylcholinesterase enzyme in the hippocampal tissue of treated rats compared to the HC group and also had a partial effect on the antioxidant levels and defense against oxidative stress in the rats receiving HC diet but this difference was not statistically significant.
