اثر هشت هفته تمرین هوازی تداومی با شدت متوسط بر شاخص های آپوپتوز بافت قلب موش های نر سالمند

چکیده

مقدمه و هدف: فرایند پیری با مجموعه‌ای از تغییرات مولکولی و سلولی در طول سال‌ها مشخص می‌شود که می‌تواند به تخریب پارامترهای فیزیولوژیکی مهم برای زنده و سالم نگه داشتن یک موجود زنده منجر شود. از طرفی فعالیت بدنی نه تنها باعث بهبود عملکرد قلبی می­شود، بلکه خطر بیماری­های قلبی را هم کاهش می­دهد. از این رو هدف این  پژوهش، بررسی اثر هشت هفته تمرین هوازی تداومی (MICT) برشاخص های آپوپتوز بافت قلب موش های نر سالمند بود.
مواد و روش ها: در این مطالعه تجربی 16 سرموش صحرایی نر از نژاد ویستار با میانگین سنی 25/2±19 ماه و وزن12/20±02/ 350 گرم به طور تصادفی به دوگروه تمرین تداومی با شدت متوسط (MT) و کنترل(C) تقسیم شدند. برنامه تمرین هوازی تداومی(MICT) به مدت 8 هفته و پنج جلسه در هفته با شدت 60-75 درصد حداکثر سرعت به صورت فزاینده انجام شد. 48 ساعت پس از آخرین جلسه تمرینی پس از بیهوشی کامل و کشتار موش ها  بافت قلب برداشته و بلافاصل در محلول نیتروژن فریز و در دمای 80- درجه نگهداری شد. شد. مقادیر بیان ژن شاخص های BAX،BCL2، نسبت BAX به BCL2، P53 وکاسپاز 3 قلب به روش Real time PCRاندازه گیری شد. داده ها  به روش آماری t مستقل در نرم افزار SPSS 27 و در سطح معنی داری (05/0>P)  تجزیه وتحلیل شدند.
نتایج: نتایج نشان داد انجام هشت هفته تمرین MICT باعث کاهش معنی دار شاخص های آپوپتوزی BAX (002/0P=) ، P53 (015/0P=) و کاسپاز3 (002/0P=)  دربافت قلب موش های سالمند نسبت به گروه کنترل شد. همچنین نتایج نشان داد اجرای تمرین به روش MICT باعث افزایش BCL2 (011/0P=)  و کاهش نسبت BAX به BCL2 (008/0P=)  در بافت قلب موش های سالمند شد.
نتیجه‌گیری: به نظر می­رسد انجام منظم تمرینات هوازی تداومی به روش MICT می­تواند در کاهش شاخص­های آپوپتوز بافت قلب و کند شدن سرعت فرآیند پیری نقش بسزایی داشته باشد.

Background and Objective: The aging process is characterized by a series of molecular and cellular changes over the years that can lead to the destruction of important physiological parameters to keep an organism alive and healthy. On the other hand, physical activity not only improves heart function but also reduces the risk of heart disease. Therefore, this study aimed to investigate the effect of eight weeks of continuous aerobic training (MICT) on the apoptosis indices of heart tissue in aged male rats.
Materials and Methods: In this experimental study, 16 male Wistar rats with an average age of 19±2.25 months and weight of 350.02±20.12 grams were used. They were randomly divided into two groups of moderate intensity continuous training (MT) and control (C). The continuous aerobic exercise program (MICT) was performed for 8 weeks and five sessions per week with an intensity of 60-75% of the maximum speed. 48 hours after the last training session, after complete anesthesia and killing of the mice, heart tissue was removed and immediately frozen in nitrogen solution and kept at -80 degrees. The gene expression values of BAX, BCL2, the ratio of BAX to BCL2 and P53, and caspase 3 of the heart were measured by real-time PCR. The data were analyzed by independent t-statistical method in SPSS 27 software at a significant level of P<0.05.
Results: The findings showed that eight weeks of MICT training significantly reduced the apoptotic indices of BAX (P=0.002), P53 (P=0.015), and caspase 3 (P=0.002) in the heart tissue of elderly rats compared to the control group. Also, the results showed that MICT training increased BCL2 (P=0.011) and decreased the ratio of BAX to BCL2 (P=0.008) in the heart tissue of elderly rats.
Conclusion: It seems that regular continuous aerobic exercise using the MICT method can play a significant role in reducing the apoptotic indices of the heart tissue and slowing down the aging process.
