بررسی تاثیر هشت هفته تمرین هوازی بر بیان ATP-binding cassette transporter در سلول های PBMC زنان میانسال یائسه

چکیده

مقدمه و هدف: بیماری آترواسکلروزیس یکی از شایع ترین علل مرگ و میر در جوامع مختلف است که در نتیجه آترواسکلروز سرخرگ های کرونر اتفاق می افتد و این عارضه در برخی از مراحل زندگی تشدید می یابد که یکی از این مراحل دوره یائسگی می باشد. در این مرحله ﺑﻪ دﻟﯿﻞ ﮐﺎﻫﺶ ﻓﯿﺰﯾﻮﻟﻮژﯾﮏ ﺳﻄﻮح اﺳﺘﺮوژن و اﯾﺠﺎد ﭼﺎﻗﯽ آﻧﺪروﺋﯿﺪ، اﻓﺰاﯾﺶ ﻓﻌﺎﻟﯿﺖ ﺳﯿﺴﺘﻢ ﺳﻤﭙﺎﺗﯿﮏ، ﻧﻘﺺ در ﻋﻤﻠﮑﺮد و اﮐﺴﯿﺪاﺳﯿﻮن ﭼﺮﺑﯽﻫﺎ، اﺧﺘﻼل درﻋﻤﻠﮑﺮد اﻧﺪوﺗﻠﯿﺎل و اﻟﺘﻬﺎب ﻋﺮوﻗﯽ اﺣﺘﻤﺎل اﺑﺘﻼ ﺑﻪ ﺑﯿﻤﺎریﻫﺎی ﻗﻠﺒﯽ-ﻋﺮوﻗﯽ را در زﻧﺎن ﯾﺎﺋﺴﻪ اﻓﺰاﯾﺶ ﻣﯽیابد. یکی از عوامل کاهش خطر آترواسکلروزیس بیان ATP-binding cassette transporter (ABCA) mRNA expression است که در انتقال معکوس کلسترول نقش دارد. هدف از مطالعه حاضر بررسی تاثیر تمرین هوازی بر بیان mRNA ABCA1 در سلول های PBMC می باشد.
مواد و روش ها: این مطالعه بر روی 30  نفر زن یائسه به صورت تصادفی در دو گروه کنترل و گروه هشت هفته تمرین هوازی انجام شد. سپس قبل و بعد از مداخله از همه آزمودنی­ها cc10 خون­گیری به عمل آمد و جدا سازی لنفوسیت­ها به روش سانتریفیوژ و تلخیص  mRNAبه وسیله دستگاه  PCRانجام و اطلاعات حاصل توسط تحلیل کوواریانس در سطح معنی­داری  (P≤0.05) و تعقیبی بونفرونی جهت بررسی بین گروه ها در محیط SPSS19 تحلیل شد.
نتایج: داده ها  نشان داد تمرین هوازی در بیان ژنABCA1 و HDL و LDLو  APO-A1تاثیر معنی­داری داشته است. (P ≤0.05).
نتیجه‌گیری: براساس یافته­های تحقیق می­توان نتیجه­گیری نمود که هشت هفته تمرین هوازی باعث بهبود انتقال معکوس کلسترول و پیشگیری از بیماری آترواسکلروزیس در زنان یائسه می شود .

Background and Objective: Atherosclerosis is one of the most common causes of death in different societies, which occurs as a result of atherosclerosis of the coronary arteries and this complication is exacerbated in some stages of life, one of which is menopause. At this stage, due to reduced physiological estrogen levels and android obesity, increased activity of the sympathetic system malfunction and fat oxidation, endothelial dysfunction and vascular inflammation risk of cardiovascular disease in postmenopausal women increases. One of the factors reducing the risk of atherosclerosis is the expression of ATP-binding cassette transporter (ABCA) 1 mRNA, which is involved in reverse cholesterol transfer. The aim of this study was to evaluate the effect of aerobic exercise on ABCA1 mRNA expression in PBMC cells.
Materials and Methods: This study was performed on 30 post-menopausal women who were randomly divided into two groups of control and eight weeks of aerobic exercise.
Then, before and after the intervention, blood samples were taken from all subjects in 10 cc and lymphocytes were isolated by centrifugation and mRNA was isolated and assessed by PCR and the information obtained were analyzed by covariance tests with significance at P <0.05 in SPSS19.
Results: The results showed that aerobic exercise had a significant effect on the expression of ABCA1, HDL, LDL and APO-A1 genes (P <0.05).
Conclusion: Based on the research findings, it can be concluded that eight weeks of aerobic exercise improves reverse cholesterol transfer and prevents atherosclerotic factors in post-menopausal women.
