بررسی اثر عصاره های جلبک Sargassum tenerrimum بر بیان ژن VEGF در رده سلولی سرطان کولورکتال SW742

چکیده

مقدمه و هدف: سرطان روده‌ی بزرگ از جمله سرطان های شایع است که مرگ‌ومیر بسیاری را در سراسر جهان موجب می‌شود. تلاش برای یافتن ترکیبات دارای پتانسیل ضد سرطانی به ویژه از محیط‌های دریایی با توجه به غنی بودن این محیط‌ها در زمینه  ی دارا بودن ارگانیسم‌های تولید کننده‌ی ترکیبات فعال زیستی، از جمله موضوعات مورد توجه محققین است. بر همین اساس هدف از تحقیق حاصل، بررسی اثر عصاره ی جلبک دریایی Sargassum tenerrimum بر بیان ژن فاکتور رشد اندوتلیال عروق (VEGF) در سلول های سرطانی کولورکتال بود.
مواد و روش ها: در تحقیق حاضر سه عصاره ی متانولی، کلروفرمی و هیدروالکلی از جلبک دریایی Sargassum tenerrimum  خلیج فارس، تهیه شد و اثر سه غلظت 250، 500 و 750 میلی  گرم بر میلی لیتر هر یک از عصاره ها بر روی بیان ژن VEGF در سلول  های SW742 رده‌ی سرطان کولورکتال با استفاده از روش Real Time PCR ارزیابی گردید. آنالیز داده‌های حاصل با استفاده از آزمون One Way ANOVA همراه با تست LSD انجام شد.
نتایج: نتایج حاصل نشان داد که عصاره  ی هیدروالکلی در غلظت 500 و 750 میلی‌گرم بر میلی  لیتر و عصاره‌های کلروفرمی و متانولی در غلظت 700 میلی‌گرم بر میلی  لیتر موجب کاهش معنادار بیان VEGF در سلول  های مورد مطالعه می  شوند (05/0P<).
نتیجه‌گیری: این نتایج پتانسیل ضد سرطانی عصاره‌ی Sargassum tenerrimum را از طریق کاهش بیان VEGF و آنژیوژنز نشان می  دهد.

Background and Objective: Colon cancer is one of the most common cancers that causes many deaths worldwide. Efforts to find compounds with anti-cancer potential, especially from marine environments due to the richness of these environments in the field of organisms producing bioactive compounds, are among the topics of interest to researchers. Accordingly, the aim of this study was to investigate the effect of Sargassum tenerrimum seaweed extract on the expression of vascular endothelial growth factor (VEGF) gene in colorectal cancer cells.
Materials and Methods: In the present study, three methanolic, chloroform and hydroalcoholic extracts of Sargassum tenerrimum seaweed from Persian Gulf were prepared and the effect of three concentrations 250, 500 and 750 mg/ml of each extract on VEGF expression was evaluated in SW742 cells of colorectal cancer using Real Time PCR. Data were analyzed using One Way ANOVA along with LSD test.
Results: The results showed that hydroalcoholic extracts at concentrations of 500 and 750 mg/ml and chloroform and methanolic extracts at concentrations of 700 mg/ml caused a significant decrease in VEGF expression in SW742 cells (P <0.05).
Conclusion: These results indicate the anti-cancer potential of Sargassum tenerrimum extract by reducing VEGF expression and angiogenesis.
