تاثیر تمرین تناوبی شدید و رژیم غذایی پرپروتئین بر شاخص های آپوپتوزی کاسپاز-3، Bax و Bcl-2 بافت قلب موش های بزرگ آزمایشگاهی نر چاق

چکیده

مقدمه و هدف: چاقی در آپوپتوز سلول های قلب و بروز بیماری های قلبی- عروقی نقش دارد. تمرینات ورزشی و اصلاح رژیم غذایی می تواند بر عوامل آپوپتوزی تاثیرگذار باشد. هدف از مطالعه حاضر بررسی اثر تمرین تناوبی شدید (HIIT) و رژیم غذایی پرپروتئین (HPD) بر شاخص های آپوپتوزی کاسپاز-3، Bax و Bcl-2 بافت قلب موش های بزرگ آزمایشگاهی چاق بود.
مواد و روش ها: در مطالعه تجربی حاضر، 40 سر موش بزرگ آزمایشگاهی نر چاق به پنج گروه مساوی شامل HIIT (شدت 90 درصد حداکثر اکسیژن مصرفی، پنج جلسه  در هفته، 10 هفته)، HPD، HIIT+HPD، کنترل چاق-1 (OC-1) و کنترل چاق-2 (OC-2) و هشت سر موش غیر چاق نیز در گروه کنترل غیرچاق (NC) تقسیم شدند. بافت قلب 48 ساعت پس از آخرین جلسه مداخله برای اندازه گیری سطوح پروتئین های کاسپاز-3، Bax و Bcl-2 خارج شد. آنالیز داده‌ها با استفاده از آزمون‌های آنالیز واریانس یک طرفه، کروسکال- والیس و من ویتنی در سطح معناداری 05/0≥P انجام شد.
نتایج: سطوح کاسپاز-3 و Bax در گروه HIIT+HPD نسبت به گروه های HIIT، HPD و OC-1 به طور معناداری پایین تر بود. از طرفی سطح Bcl-2 در گروه های HIIT و HPD نسبت به گروه های OC-1 و NC به طور معناداری بالاتر بود. همچنین نسبت Bcl-2 به Bax در گروه های HIIT و HIIT+HPD در مقایسه با گروه OC-1 به طور معناداری بالاتر بود.
نتیجه گیری: با توجه به نتایج مطالعه حاضر، HIIT به همراه HPD باعث بهبود وضعیت آپوپتوزی بافت قلب می شود. به نظر می رسد HIIT به همراه HPD، سازگاری های لازم برای مهار آپوپتوز ناشی از چاقی را در بافت قلب موش های بزرگ آزمایشگاهی فراهم می سازد.

چکیده [English]

Background and Objective: Obesity is involved in the apoptosis of cardiac cells and the development of cardiovascular diseases. Exercise training and diet modification can affect apoptotic indices. The aim of the present study was to assess the effect of high-intensity interval training (HIIT) and high-protein diet (HPD) on the apoptotic indices of caspase-3, Bax, and Bcl-2 in the cardiac tissue of obese rats.
Materials and Methods: In the present experimental study, 40 obese male rats were divided into five equal groups, including HIIT (90% VO2max, 5 sessions per week, 10 weeks), HPD, HIIT+HPD, obesity control-1 (OC-1), obesity control-2 (OC-2), and eight non-obese rats were also considered in the non-obese control (NC) group. Cardiac tissue was extracted 48 hours after the last intervention session to measure the levels of caspase-3, Bax, and Bcl-2 proteins. Data were analyzed using one-way ANOVA, Kruskal-Wallis, and Mann-Whitney tests (P≤0.05).
Results: The caspase-3 and Bax levels in the HIIT+HPD group was significantly lower than in the HIIT, HPD, and OC-1 groups. On the other hand, the HIIT and HPD groups showed a significantly higher Bcl-2 level than the OC-1 and NC groups. Also, there was a significantly higher of Bcl-2 to Bax ratio in the HIIT and HIIT+HPD groups compared with the OC-1 group.
Conclusion: According to the results of the present study, a combination of HIIT and HPD improves the apoptotic status of cardiac tissue. It seems that the combination of HIIT and HPD provides the necessary adaptations to inhibit obesity-induced apoptosis in the cardiac tissue of obese rats.
