تأثیر گزنه همراه با تمرین استقامتی بر شاخص‌های اسپرماتوژنز در بیضه موش‌های سفید بزرگ آزمایشگاهی دیابتی‌شده توسط استرپتوزوتوسین

چکیده

مقدمه و هدف: امروزه دیابت یک مشکل بهداشتی بزرگ با شیوع فزاینده در مردان است. بیان شده است که گلوکز بالا منجر به افزایش استرس اکسیداتیو و آپوپتوز در سلول‎های بیضه می‌شود و باعث مرگ سلول‌های بیضه و ناباروری نیز می‌شود. این مطالعه به بررسی و تعیین اثر عصاره هیدروالکلی گزنه و تمرینات استقامتی بر ضرایب اسپرماتوژنز در موش‌های سفید بزرگ نر دیابتی‌شده توسط STZ انجام شد.
مواد و روش ها: در این مطالعه تجربی، 40 سر موش سفید بزرگ آزمایشگاهی با وزن 200-220 گرم به‌صورت تصادفی به 5 گروه کنترل-سالم (HC)، گزنه-دیابت (DG)، تمرین-دیابت (DT)، گزنه-تمرین-دیابت ((DTG) و کنترل-دیابت (DC) تقسیم شدند. تمرین استقامتی در 6 هفته طبق پروتکل اجرا شد. به‌طور هم‌زمان، موش‌های سفید بزرگ آزمایشگاهی گروه‌های DG و DTG عصاره هیدروالکلی گزنه را به میزان 45 میلی‌گرم به ازای هر کیلوگرم وزن بدن دریافت کردند. بعد از 6 هفته بیضه‌ها برداشته و با تکنیک H & E رنگ‌آمیزی شدند.
نتایج: در گروه DTG و گروه DG، هر دو شاخص SI (spermatogenesis index) و RI (repopulation index) در مقایسه با گروه DT بالاتر بود و به نتایج گروه HC نزدیک بود. همچنین بررسی‌های حاصل از هیستوپاتولوژی بیضه در این دو گروه بهبود بالاتری در مقایسه با گروه DT نشان دادند و نتایج مشابهی با نتایج گروه HC به‌دست آمد.
نتیجه‌گیری: مطالعه حاضر نشان داد که دیابت اثرات مخربی بر اسپرماتوژنز و بافت بیضه دارد و تجویز گزنه و تمرینات استقامتی همراه با یکدیگر می‌تواند با بهبود درصد SI و RI، سلول‌های اسپرماتوژنیک در لوله‌های سمی‌نیفر را از اثرات مضر دیابت محافظت کند.

Background and Objective: Today, diabetes is a major health problem with an increasing prevalence in men. It has been stated that high glucose leads to increased oxidative stress and apoptosis in testicular cells and causes testicular cell death and infertility. This study was conducted to investigate and determine the effect of nettle hydroalcoholic extract and endurance exercises on testicular tissue and spermatogenesis factors in streptozotocin (STZ)-diabetic male rats.
Materials and Methods: In this experimental study, 40 albino rats weighing 200-220 grams were randomly divided into 5 groups: control-healthy (HC), nettle-diabetes (DG), exercise-diabetes (DT, nettle-exercise), diabetes (DTG), and control diabetes (DC). Endurance training was performed for 6 weeks according to the protocol. At the same time, rats of DG and DTG groups received nettle hydroalcoholic extract at a dose of 45 mg per kg of body weight. After 6 weeks, the testes were removed and stained with H&E technique.
Results: In the DTG and the DG groups, both spermatogenesis index (SI) and repopulation index (RI) were higher compared to the DT group and were close to the results of the healthy control group. Also, the histopathology of the testis in these two groups showed a higher improvement compared to the DT group and similar results were obtained with the results of the HC group.
Conclusion: The present study showed that diabetes has destructive effects on spermatogenesis and testicular tissue and administration of nettle and endurance exercises together can protect spermatogenic cells in seminiferous tubules from the harmful effects of diabetes by improving SI and RI percentages.
