اثر بذر کدو و تمرین استقامتی بر نشانگان فشار اکسیداتیو و تخریب DNA

چکیده

مقدمه و هدف: این پژوهش با هدف بررسی نقش بذر کدو و تمرین استقامتی بر نشانگان فشار اکسیداتیو و تخریب DNA بافت عضلات کند انقباض در موش های صحراییی نر مسموم شده با پراکسید هیدروژن انجام شد.
مواد و روش ها: تعداد 48 سر موش صحرایی نر بالغ با وزن 20±220 گرم و 8 هفته ، مطابق با مداخلات استرس (پراکسیدهیدروژن )، مکمل بذر کدو و تمرین استقامتی اجرا شد. جهت القا استرس اکسیداتیو تزریق درون صفاقی H2O2 با دوزmmol/kg  2به صوموش صحرایی سه بار در هفته‌یک روز در میان انجام شد. القا مکمل بذر کدو به صوموش صحرایی 5/0 میکروگرم روزانه با تزریق درون صفاقی با غلظت  UI/ml 300000 انجام شد. گروه های تمرینی روزانه تمرین هوازی بر روی تردمیل را به مدت 8هفته انجام دادند، پروتکل تمرین با سرعت m/min  8 و شیب 10 درجه به مدت 30 دقیقه بر روی تردمیل در هفته‌ی اول شروع و به تدریج به سرعت m/min  20با زاویه ده درجه به مدت 60 دقیقه در هفته‌ی هشتم رسید. جهت تجزیه تحلیل داده‌ها از آزمون تحلیل واریانس با اندازه‌گیری مکرر و آزمون تعقیبی بونفرونی در سطح 05/0=  α استفاده گردید.
نتایج: نتایج پژوهش حاضر نشان داد که تمرین و دریافت بذر کدو به صوموش صحرایی جداگانه اثر معنی داری بر غلظت ATP، ADP، نسبت ATP/ADP ، MDA و PAB عضله نعلی دارد (001/0P=)؛ اما تعامل تمرین و بذر کدو فقط بر MDA عضله نعلی اثر معنی داری نداشت (025/0P=)  ولی بر سایر متغیرها تأثیر معناداری داشت (001/0P=).
نتیجه‌گیری: مصرف مکمل بذر کدو به همراه تمرین استقامتی می تواند باعث کاهش استرس اکسیداتیو در بدن گردد.

چکیده [English]

Objective: This study aimed to investigate the effect of pumpkin seed and endurance training on oxidative stress factors and DNA damage of slow-twitch muscle tissue in male rats poisoned with hydrogen peroxide.
Materials and Methods: A total of 48 adult male rats weighing 220±20 g at eight weeks were administered in accordance with stress interventions (hydrogen peroxide), pumpkin seed and endurance training. To induce oxidative stress, intraperitoneal injection of H2O2 at a dose of 2 mmol/kg was performed three times a week. Inoculation of pumpkin seeds was performed at 0.5 μg per day with intraperitoneal injection at a concentration of 300000 U/ml. The training groups performed daily aerobic exercise on a treadmill for 8 weeks, the endurance training protocol started at a speed of 8 m/min and a slope of 10 degrees for 30 minutes on the treadmill in the first week and gradually at a speed of 20 m/min at an angle of ten. The degree reached 60 minutes in the eighth week. Data were analyzed using repeated- measures analysis of variance and Bonferroni post hoc test at the level of α = 0.05.
Results: The results of the present study showed that training and receiving pumpkin seeds separately has a significant effect on ATP, ADP, ATP / ADP ratio, MDA and PAB of horseshoe muscle (P= 0.001); But the interaction between training and pumpkin seeds did not have a significant effect only on MDA of soleus muscle (P = 0.025) but had a significant effect on other variables (P = 0.001).
Conclusion: Taking pumpkin seed supplement along with endurance training can reduce oxidative stress in the body.
