اثر تمرین منظم شنا و عصاره سیر بر مرگ برنامه ریزی شده سلول های کبدی در موش‌های پیر تحت القا دوکسوروبیسین

چکیده

مقدمه و هدف: عامل اصلی بیماری‌های وابسته به سن را می‌توان افزایش آسیب اکسایشی DNA ذکر کرد. این آسیب می‌تواند منجر به ترجمان ژن‌های خاصی شود که تکثیر و مرگ سلولی را تنظیم می‌کنند. هدف از مطالعۀ حاضر، بررسی اثر یک دوره تمرین منظم شنا به‌همراه مصرف عصارۀ سیر بر میزان Bax و Bcl-2 بافت کبد موش‌های پیر تحت‌القای دوکسوروبیسین می‌باشد.
 
مواد و روش‌ها: چهل‌ودو سر موش صحرایی نر مسن 40 تا 50 هفته‌ای با میانگین وزن اولیۀ 250 تا 300 به‌روش تصادفی به شش گروه تقسیم شدند: 1. کنترل؛ 2. سالین؛ 3. دوکسوروبیسین؛ 4. دوکسوروبیسین و تمرین؛ 5. دوکسوروبیسین و سیر؛ 6. دوکسوروبیسین و سیر و تمرین. برای القای بیماری مزمن کلیوی از داروی دوکسوروبیسین، به‌میزان 5/8 میلی‌لیتر به‌ازای هر کیلوگرم وزن استفاده شد. برنامۀ تمرینی اصلی به‌صورت تمرین شنا به‌مدت هشت هفته و هر هفته سه روز و هر روز سی دقیقه بود. گروه‌های دریافت‌کنندۀ مکمل و تمرین و مکمل، روزانه یک میلی‌لیتر عصارۀ سیر به‌ازای هر کیلوگرم وزن بدن، به‌مدت هشت هفته به‌صورت خوراکی (گاواژ) دریافت کردند. چهل‌وهشت ساعت پس از آخرین جلسۀ تمرینی و پس از 10 تا 12 ساعت ناشتایی، موش‌ها بی‌هوش شده و بافت کبد بلافاصله جدا و در فریزر با دمای منفی هفتاد درجۀ سانتی‌گراد برای اندازه‌گیری سطوح Bax و Bcl-2 استفاده گردید. تجزیه‌وتحلیل داده‌ها با استفاده از آزمون آنالیز واریانس یک‌طرفه و آزمون تعقیبی توکی جهت تعیین محل اختلاف بین گروه‌‌ها استفاده گردید.
 
نتایج: نتایج نشان داد هشت هفته تمرین شنا و مصرف عصارۀ سیر و ترکیب تمرین و سیر سبب افزایش معنی‌داری در سطوح Bcl-2 و همچنین کاهش معنی‌‌داری در سطوح Bax و نسبت Bcl-2/Bax بافت کبد موش‌های مسن تحت‌القای دوکسوروبیسین شد (05/0>P < /span>).
 
نتیجه‌گیری: فعالیت ورزشی، استفاده از مکمل عصارۀ سیر و ترکیبی از این دو شیوه می‌توانند، به‌عنوان روش‌های درمانی غیردارویی مؤثر، جهت کاهش آسیب کبدی و حمایت بافت کبد در مقابل آسیب‌های ناشی از استرس اکسایشی و التهابی افزایش سن محسوب شوند

چکیده [English]

Background and Objective: Increased oxidative damage to DNA can be mentioned as the main cause of age-related diseases. This damage can result in translation of specific genes which regulate cell proliferation and apoptosis. The aim of this study was to investigate the effect of a regular swimming training with garlic extract on the levels of Bax and Bcl-2 in the liver tissue doxorubicin -induced aging rat
 
Materials and Methods: In this study, 42 aged male rats, 40-50 weeks, with an average initial weight of 250-300 g were randomly divided into 6 groups: 1. control, 2. saline, 3. doxorubicin, 4. doxorubicin+exercise, 5. doxorubicin+garlic, and 6. doxorubicin+garlic+exercise. Chronic kidney disease (CKD) was induced by injection of 8.5 ml/kg of doxorubicin. The animals in the exercise groups were forced to swim 30 min once a day, 3 days per week, for 8 weeks. Groups receiving supplement and exercise + supplement, daily treated with 1 ml of garlic extract per kilogram of body weight for 8 weeks orally (gavage). In addition, 48 hours after the last training session and after 12-10 hours of fasting, rats were anesthetized and liver tissue separated immediately and stored at -70ᵒC to measure the levels of Bax and Bcl-2. For comparisons among the groups, one-way analysis of variance (ANOVA) and Tukey’s test were performed with P < /em><0.05 as an indication of statistical significance.
 
Results: The findings showed that eight weeks of swimming training and garlic extract and the combination of exercise training and garlic extract resulted in a significant increase in the levels of Bcl-2 and also a significant decrease in the levels of Bax and Bax/Bcl-2 in the kidney tissue of aged rats induced by doxorubicin (P<0.05).
 
Conclusion: Exercise training, herbal supplement of garlic, and a combination of these two methods can be as an effective non-pharmacological therapy to reduce liver damage and protect liver tissue against oxidative stress and inflammatory injuries associated with aging.
