اثر عصاره آبی الکلی انیسون بر استرس اکسیداتیو و فعالیت بوتیریل کولین استراز هیپوکمپ در موش‌های سفید بزرگ آزمایشگاهی دریافت‌کننده رژیم پرکلسترول

چکیده

مقدمه و هدف: مصرف رژیم غذایی پرکلسترول و استرس اکسیداتیو ناشی از آن موجب بروز انواع بیماری‌های عصبی می‌شود. با توجه به خواص ضدالتهابی و آنتی‌اکسیدانی گیاه انیسون، در این مطالعه اثر عصاره آبی-الکلی این گیاه بر استرس اکسیداتیو و فعالیت کولین استراز هیپوکمپ موش‌های سفید بزرگ آزمایشگاهی دریافت‌کننده رژیم پرکلسترول بررسی شد.
مواد و روش‌ها: در این مطالعه تجربی، موش‌ها به چهار گروه کنترل، کنترل دریافت‌کننده عصاره، پرکلسترول، و پرکلسترول و دریافت‌کننده عصاره تقسیم شدند. پس از 8 هفته، گروه‌های تحت درمان، با عصاره انیسون به میزان mg/kg 250به‌صورت داخل صفاقی به مدت 3 هفته درمان شدند و هم‌زمان تجویز غذای پرکسترول تا پایان مطالعه ادامه داشت. در پایان کار، پس از تهیه هموژنه هیپوکمپ، سنجش فعالیت بوتیریل‌کولین‌استراز (BChE) و شاخص‌های استرس اکسیداتیوشامل رادیکال‌های آزاد اکسیژن (ROS)، گلوتاتیون (GSH) و کاتالاز انجام شد. داده‌ها از نظر آماری با آزمون آنووای یک‌طرفه و تست تعقیبی توکی آنالیز شدند.
نتایج: رژیم غذایی پرکلسترول باعث افزایش معنا‌دار فعالیت BChE و ROS و همچنین کاهش معنا‌دار GSH و کاتالاز هیپوکمپ شد. از طرف دیگر، تیمار گروه پرچرب با عصاره انیسون موجب کاهش معنا‌دار ROS و BChE و افزایش معنا‌دار GSH شد و بر فعالیت کاتالاز تأثیر معنادار نداشت.
نتیجه‌گیری: مصرف عصاره آبی-الکلی انیسون موجب کاهش فعالیت ‌کولین‌استراز و استرس اکسیداتیو هیپوکمپ به دنبال دریافت رژیم پرکلسترول می‌شود که این احتمالاً در کاهش عوارض عصبی مرتبط می‌تواند سودمند باشد.

Background and Objective: High-cholesterol diet and ensuing oxidative stress causes development of various neurologic disorders. Considering anti-inflammatory and antioxidant properties of the anise (Pimpinella anisum) plant, the effect of its hydroalcoholic extract on hippocampal oxidative stress and cholinesterase activity was assessed in rats under high-cholesterol diet.
Materials and Methods: In this experimental study, rats were divided into control, extract-treated control, high cholesterol diet, and extract-treated high cholesterol diet. After 8 weeks, treated groups also received extract at a dose of 250 mg/kg (i.p.) for 3 weeks and high-cholesterol diet was given till the end of the study. Then, after preparing hippocampal homogenate, butyrylcholinesterase (BChE) activity and oxidative stress factors including reactive oxygen species (ROS), glutathione (GSH), and catalase activity were determined. One-way ANOVA and Tukey's post-hoc tests were used for statistical analysis of data.
Results: High cholesterol diet significantly increased hippocampal BChE activity and ROS level and significantly decreased GSH and catalase activity. Conversely, extract treatment of group under high-cholesterol diet caused significant decrease of ROS and BChE activity and significant increase of GSH and with no significant effect on catalase activity.
Conclusion: Consumption of anise hydroalcoholic extract under high-cholesterol diet reduces hippocampal oxidative stress and cholinesterase activity and this may lower associated neurological complications.
