بررسی اثرات توأم ورزش فکری و نپتا منتوییدس ( اسطوخودوس خراسانی ) بر افسردگی ناشی از رزرپین در موش بزرگ آزمایشگاهی نر

چکیده

مقدمه و هدف: مطالعات نشان داده اند که تقویت قوای ذهنی ابتلا به افسردگی را کاهش می دهد. گزارش هایی از خواص ضد افسردگی گیاه نپتا منتوییدس نیز موجود می باشد. هدف از این مطالعه سنجش اثر ورزش ذهنی و نپتا منتوییدس و نیز مقایسه اثر آن دو بر افسردگی ناشی از رزرپین در موش های بزرگ آزمایشگاهی می باشد.
مواد و روش ها: موش های نر به 8 گروه تقسیم شدند: 1- کنترل 2- رزرپین 3- رزرپین+ فلوکستین 4- نپتا 5- نپتا+رزرپین6- ورزش ذهنی 7- ورزش ذهنی+رزرپین 8 - ورزش ذهنی+ نپتا+رزرپین. رزرپین با دز 2/0 (mg/kg) در طول 14 روز  داخل صفاقی تزریق شد . همزمان موش ها در ماز شعاعی هشت بازویی آموزش گرفته و عصاره نپتا را با دز 200 (mg/kg) دریافت کردند . سپس نتایج حاصل از تست های شنای اجباری، ترجیح سوکروز، میدان باز، ماز بعلاوه مرتفع با آنالیز واریانس یکطرفه  ارزیابی شدند.
یافته ها: رزرپین زمان بی حرکتی را افزایش و زمان شنای فعال را کاهش داد، در حالی که ترکیب اثر ورزش فکری و نپتا بر اثر رزرپین غلبه کرد. رزرپین درصد استفاده از سوکروز را کاهش داد که این اثر توسط درمان با ورزش ذهنی، نپتا و ترکیب آنها بهبود یافت. کاهش فعالیت لوکوموتور ناشی از رزرپین در گروه های ورزش ذهنی و نپتا بهبود یافت. رزرپین زمان ماندن در بازوی باز را کاهش داد و نپتا آن را جبران کرد.
نتیجه گیری: ورزش ذهنی و نپتا علایم رفتاری افسردگی و اضطرب ناشی از رزرپین را بهتر از  فلوکستین بهبود بخشیدند.

Background and Objective: Many studies demonstrate that cognitive empowerment reduces the risk of depression. Anti-depressant properties of Nepeta menthoides has also been reported. This study aimed to determine the effects of mental exercise and Nepeta menthoides and also to compare both of them on reserpine-induced depression in rats.
Materials and Methods: Male rats were divided into 8 groups: 1- Saline, 2- Reserpine, 3-Reserpine + Fluoxetine, 4- Nepeta, 5- Nepeta +Reserpine, 6- Mental exercise, 7- Mental exercise+ Reserpine, 8- Mental exercise + Nepeta + Reserpine. Reserpine was administered intraperitoneally at a dose of 0.2 mg/kg for 14 days. At the same time, the animals received Nepeta at a dose of 200 mg/kg and radial maze training was conducted. Then, the results for forced swimming test (FST), sucrose preference (SPT), elevated plus maze (EPMT) and open field tests (OFT) were assessed through one-way Analysis of Variance.
Results: Reserpine significantly enhanced immobility and decreased active swimming time, while mental exercise combined with Nepeta were able to antagonize the effect of reserpine. Reserpine also lowered sucrose preference ratio which were recovered in mental exercise, Nepeta and combined treatment groups. Decreased locomotor activity due to reserpine was improved in mental exercise and Nepeta groups. Time spent and % of entrance into the open arms reduced by reserpine and Nepeta compensate it.
Conclusion: Mental exercise and Nepeta menthoides improve the depression and behavioral symptoms of anxiety induced by reserpine better than fluoxetine.
