اثر عصاره آبی الکلی اسطوخودوس رومی (Lavandula dentata) بر یادگیری و حافظه در موش‌های نر دیابتی شده با استرپتوزوتوسین

چکیده

مقدمه و هدف: دیابت یک اختلال متابولیک مزمن است و استرس اکسیداتیو نقش مهمی در ایجاد عوارض دیابت مانند اختلال حافظه و یادگیری ایفا می‌کند. از آنجا که گونه‌های لوندولا موجب بهبود حافظه و یادگیری می‌گردند و Lavandula dentata دارای اثرات آنتی‌اکسیدانی و ضدالتهابی است و در ایران تحت عنوان اسطوخودوس استفاده می‌شود بر آن شدیم تا اثرات آن را بر اختلال یادگیری و حافظه ناشی از دیابت مطالعه کنیم.
 
مواد و روش‌ها: موش‌های صحرایی نر به تعداد 40 سر به 4 گروه کنترل، کنترل دیابتی، سالم با لوندولا دنتاتا، دیابتی با لوندولا دنتاتا تقسیم گردیدند. برای دیابتی نمودن حیوانات از تزریق داخل صفاقی تک‌دوز استرپتوزوتوسین استفاده شد. پس از 6 هفته، درمان با لوندولا دنتاتا به فرم تزریق داخل صفاقی با دوز 400 میلی‌گرم بر کیلوگرم به مدت دو هفته نیم ساعت قبل از انجام تست‌های رفتاری صورت گرفت. جهت ارزیابی حافظه و یادگیری از ماز شعاعی هشت بازویی و تست آماری Two way repeated –measures ANOVA و در سنجش قند از آنوای یک‌طرفه استفاده شد.
 
نتایج: دیابت موجب آسیب یادگیری و حافظه به شکل افزایش خطاهای حافظه کاری و رفرنس گردید و مصرف لوندولا در گروه دیابتی سبب کاهش معنی‌دار تعداد خطاهای مربوطه گردید. اسطوخودوس رومی همچنین موجب کاهش قند خون در گروه دیابتی شد.
 
نتیجه ­گیری: مصرف لوندولا دنتاتا در موش‌های دیابتی سبب بهبود اختلال حافظه و یادگیری ناشی از دیابت شد. این اثر می‌تواند حداقل بخشی ناشی از کاهش قند خون باشد.

Background and Objective: Diabetes is a chronic metabolic disorder and oxidative stress plays an important role in the development of diabetic complications such as learning and memory impairment. Since Lavendula species improve learning and memory and Lavandula dentata contains antioxidant and anti-inflammatory effects and it is used as ustokhoddus in Iran, we have designed this research to study its effects on diabetes-induced learning and memory impairment.
 
Materials and Methods: Forty male rats were divided into 4 groups including control, control diabetic, healthy Lavendula dentata, and diabetic Lavendula dentata. A single dose of streptozotocin was used for diabetes induction in diabetic animals. After 6 weeks, Lavendula dentata treatment was performed as an intraperitoneal injection of 400 mg/kg for two weeks, 30 min before each behavioral test. The eight-arm radial maze method with two way repeated-measures ANOVA test were used to evaluate learning and memory and one-way ANOVA was used for comparing blood glucose between different groups.
 
Results: Diabetes caused learning and memory damage in the form of increasing working and reference memory errors and use of Lavendula dentata in the diabetic group caused a significant reduction in the number of related errors. Romian ustokhoddus also declined blood glucose in diabetic group.
 
Conclusion: Lavendula dentata consumption in diabetic rats improved diabetes-induced learning and memory impairment. This effect might be mediate at least partly by decreasing blood glucose.
