بررسی تداخل اثر سم عقرب مزوبوتوس اپیوس و داروی سیکلوسپورین در مکانیسم‌های بیوشیمیایی و هیستولوژی در بافت پارانشیم کبد و کلیه موش سوری

چکیده

مقدمه و هدف: زهر  Mesobuthus به عنوان خانواده buthidae شناخته می‌شود که بر اساس مطالعات انجام شده، این زهر می‌تواند باعث مرگ انسان شود. از طرفی خواص ضدالتهابی سیکلوسپورین در پیوند اعضاء از طریق سرکوب سیستم ایمنی و التهاب نشان داده شده است و احتمالاً می‌تواند با اثر زهر مقابله کند؛ بنابراین در مطالعه حاضر اثر سیکلوسپورین بر روی زهر epeus Mesobuthus  مورد بررسی قرار گرفت.
 
مواد و روش‌ها: این یک مطالعه تجربی است. تعداد 50 موش سوری به 5 گروه 10 تایی طبقه‌بندی شدند. زهر Mesobuthus epeus  و سیکلوسپورین در دوزهای10، 20 و 30 میلی‌گرمی انتخاب شد. در این مطالعه موش‌های هایی که سیکلوسپورین را دریافت نکردند به عنوان گروه کنترل در نظر گرفته شدند. پارامترهای بیوشیمیایی همراه با نیتریک اکساید و سیتوکاین  IL-2 در همه‌ی گروه‌ها بوسیله متد روتین اسپکتروفوتومتری و الایزا به ترتیب اندازه‌گیری شد.  ضمن اینکه، بررسی‌های پاتولوژیک کلیه و کبد نیز انجام شد.
 
نتایج: همه‌ی شاخص‌های بیوشیمیایی و IL-2 بطور قابل‌ملاحظه‌ای پس از تزریق زهر  Mesobuthus افزایش یافت.(P<0.001)  از سوی دیگر، گروه‌های موش پس از دریافت سیکلوسپورین کاهش معنی‌داری در همه‌ی پارامترهای مورد مطالعه نشان داند.(P<0.001).
 
نتیجه‌گیری: سیکلوسپورین در دوز 30 میلی‌گرمی قادر به کاهش پاسخ التهابی شده و می‌تواند داروی درمانی مناسبی برای بیماران عقرب‌گزیده باشد.

Background and Objective: Venom of Mesobuthus in the family buthidae can result in death in humans according to the performed studies. On the other hand, it has been demonstrated that due to anti-inflammatory properties of cyclosporine in organ transplantation through suppression of the immune system and inflammation, it could counteract venom effect. Therefore, in this study, the effect of cyclosporine on Mesobuthus eupeus venom was evaluated.
 
Materials and Methods: This was an experimental study. For this purpose, fifty mice were classified into five groups, each group consisting of ten mice. Mesobuthus venom and cyclosporine were used at three doses of 10, 20, and 30 mg. In this study, the mice which did not receive cyclosporine were considered as control group. Biochemical features plus nitric oxide and IL-2 cytokine were measured in all groups using routine methods (i.e. spectrophotometer and ELISA assay, respectively). Also, pathological examination of liver and kidney was performed.
 
Results: All the studied biochemical and IL-2 levels were significantly elevated after mesobuthus venom injection On the other hand, groups which received cyclosporine showed a significant decrease in all the studied parameters (P<0.001).
 
Conclusion: Cyclosporine at a dose of 30 mg is able to decrease inflammatory responses and it could be a suitable therapeutic drug in patients who are bitten by scorpion sting.
