بررسی اثر تزریق داخل صفاقی عصارۀ هیدروالکلی دارچین (Cinnamomum Zeylanicum) بر یادگیری اجتنابی غیرفعال در موشهای نر آلزایمریشده توسط استرپتوزوتوسین
مقدمه و هدف: بیماری آلزایمر یکی از شایعترین بیماریهای نورودژنراتیو میباشد که در آن نورونها، بهویژه در نواحی مرتبط با اعمال شناختی، موردتخریب قرار میگیرند. این بیماری علت مهم دمانس پیری است که با تخریب نورونی و زوال شناختی، بهویژه در سنین پیری مشخص میشود. در گذشته، درمانهای دارویی زیادی برای بیماری آلزایمر معرفی شدهاند که علیرغم ایجاد تحول زیاد، بهطور کامل موفق نبودهاند. ازاینرو در این تحقیق، تأثیر تزریق داخل صفاقی عصارۀ دارچین بر مرحلۀ تثبیت حافظه در موشهای صحرایی آلزایمریشدۀ نر نژاد ویستار موردبررسی قرار گرفته است.
مواد و روشها: تعداد ۳۰ سر موش صحرایی نر نژاد ویستار در ۵ گروه تقسیم شدند: ۱. گروه دستنخورده (Intact)؛ ۲. گروه کنترل دی متیل سولفوکسید (DMSO)؛ ۳. گروه شاهد دی متیل سولفوکسید ـ مایع مغزینخاعی(CSF)؛ ۴. گروه دریافتکنندۀ دی متیل سولفوکسید و استرپتوزوتوسین (با دُز mg/kg ۵/۱)؛ ۵. گروه دریافتکنندۀ استرپتوزوتوسین (STZ) بهصورت داخل بطن مغزی و عصارۀ دارچین با دُز mg/kg ۱۲۵ بهصورت داخل صفاقی. جهت ایجاد مدل آلزایمری داروی استرپتوزوتوسین به درون بطنهای مغزی تزریق گردید و پس از آن، به مدت ۴ روز موشهای صحرایی آلزایمری تحت درمان با عصارۀ دارچین بهصورت داخل صفاقی قرار گرفتند. جهت ارزیابی، یادگیری و حافظه در موشهای صحرایی از طریق آزمون یادگیری اجتنابی غیرفعال بررسی شد.
نتایج: تزریق داخل بطنی استرپتوزوتوسین موجب آسیب شدید یادگیری و حافظه در موشهای صحرایی شد و عصاره دارچین سبب بهبود یادگیری در حیوانات آلزایمری گردید (P<0.001).
نتیجهگیری: نتایج کلی این پروژه نشان داد که عصارۀ دارچین منجربه درمان ضایعات شناختی در بیماری آلزایمر میشود.

آگاهی، نگرش و عملکرد مادران در زمینۀ بهداشت دهان و دندان کودکان یک تا شش سال (مناطق 3 و 16 تهران در سال 1393)
مقدمه و ﻫﺪف: کودکان ارزشها و نگرشها نسبتبه مباحث بهداشتی را از والدینشان بهخصوص مادرانشان یاد میگیرند. آگاهی از دانش، نگرش و عملکرد مادران کودکان نقش کلیدی در ایجاد نگرش و عادات در کودکان دارد. لذا کسب اطلاعات از آگاهی مادران میتواند مبنایی برای برنامهریزیهای بهداشتی باشد. هدف این مطالعه بررسی دانش و نگرش و عملکرد مادران کودکان 1تا6 ساله میباشد.
مواد و روشها: این مطالعه توصیفیمقطعی میباشد و بر روی 450 نفر مادران کودکان 1تا6 سالۀ مراجعهکننده به مهدکودکهای مناطق 3 و 16 تهران در سال 1393 انجام گردید. نمونهگیری بهصورت تصادفی خوشهای بود. از پرسشنامۀ استاندارد KAP-WHO استفاده شد. آگاهی، نگرش و عملکرد به سه سطح ضعیف، متوسط و خوب تقسیم گردید. روایی، پایایی و پرسشنامه در مطالعات قبلی موردسنجش قرار گرفته است. دادهها با استفاده از نرمافزار آماری SPSS و آزمونهای آماری تی، ANOVA، ضریب همبستگی پیرسون و اسپیرمن مورد تجزیهوتحلیل قرار گرفت.
نتایج: در حدود 34/47درصد مادران آگاهی متوسط، 12/51درصد مادران نگرش متوسط و 23/46درصد مادران عملکرد متوسط داشتند. در کل، آگاهی و نگرش و عملکرد با سطح متوسط ارزیابی شد (05/0p<). آزمونهای آماری بیانگر آن بود که بین آگاهی، نگرش و عملکرد مادران با سن، تحصیلات، وضعیت اشتغال و وضعیت اقتصادی ارتباط وجود داشت (001/0p<).
نتیجهگیری: نتایج مطالعه نشان داد که دانش و عملکرد مادران در مراقبت از دهان و دندان کودکان ناکافی بود. لذا تدوین برنامههای آموزشی یک ضرورت است.

تأثیر هشت هفته تمرینات منتخب تایچی بر تعادل ایستا و پویای زنان مبتلا به مالتیپلاسکلروزیس با تأکید بر تیپ بدنی مزومورف و اندومورف (تحقیق کارآزمایی بالینی)
مقدمه و هدف: تیپ بدنی متفاوت عاملی برای میزان تأثیرگذاری برنامههای تمرینی بر افراد است. این مطالعه با هدف تأثیر هشت هفته تمرینات تایچی بر تعادل ایستا و پویای بیماران مبتلا به مالتیپلاسکلروزیس با تیپ بدنی مزومورف و اندومورف انجام شد.
مواد و روشها: در یک مطالعۀ کارآزمایی بالینی، 48 بیمار مبتلا به مالتیپلاسکلروزیس بهصورت هدفمند انتخاب شدند. سپس بیماران بهصورت تصادفی به دو گروه 24نفری (گروه تایچی و گروه کنترل) با دو زیرگروه 12نفری با تیپ بدنی مزومورف و اندومورف تقسیم شدند. برای برآورد تعادل آزمودنیها دستگاه تعادلسنج بایودکس استفاده شد. سپس آزمودنیهای گروه تجربی بهمدت 8 هفته تمرینات تای را انجام دادند. برای تجزیهوتحلیل نتایج از روش تحلیل کوواریانس چندمتغیری (مانکوا) در سطح معنیداری 05/ 0P≤ استفاده شد.
نتایج: بین میزان تعادل ایستا و تعادل پویای گروه کنترل با تجربی تفاوت معنادار مشاهده شد. بهبود وضعیت تعادل ایستای تیپ بدنی اندومورف بیشتر از تیپ بدنی مزومورف در گروه تمرینات تایچی مشاهده شد که به معنای اثر بخشبودن بیشتر این تمرینات بر روی تعادل تیپ بدنی اندومورف میباشد؛ درحالیکه علیرغم بهبود بیشتر تعادل پویای تیپ اندومورف نسبتبه مزومورف در گروه تمرینات تایچی، تفاوت معناداری بین تیپ اندومورف و مزومورف در پسآزمون دیده نشد.
نتیجهگیری: با توجه به نتایج تحقیق حاضر تمرینات تایچی بهعنوان یک روش مداخلهایی تأثیرگذار باعث بهبود تعادل ایستا و پویای بیماران مبتلا به اماس با تیپ بدنی اندومورف و مزومورف میشود؛ درحالیکه تأثیر این تمرینات بر تیپ بدنی اندومورف بیشتر بوده است.

بررسی درصد سلولهای کشندۀ طبیعی شبه لنفوسیت T در خون محیطی مواجههیافتگان با سولفور موستارد (مدتزمان طولانی بعد از مواجهه)
مقدمه و هدف: سلولهای کشندۀ طبیعی شبه T (NKT) جمعیتی از سلولهای T هستند که ویژگیهایی از سلولهای NK را بروز میدهند. این سلولها علاوهبر اعمال اجرایی نقش مهمی در تنظیم پاسخهای ایمنی دارند. نقش این سلولها در افراد مواجههیافته با سولفورموستارد بهخوبی روشن نشده است. هدف این مطالعه بررسی درصد سلولهای NKT در خون محیطی افراد مواجههیافته نسبتبه کنترل و ارتباط آن با عوارض ریوی میباشد.
مواد و روشها: از 77 فرد مواجههیافته با سولفورموستارد با عوارض ریوی و 62 داوطلب سالم بدون هیچ شواهد کلینیکی بهعنوان کنترل دعوت شد. اسپیرومتری انجام شد و شرکتکنندگان براساس طبقهبندی ابتکار جهانی برای بیماری انسدادی مزمن ریه (GOLD) به چهار گروه نرمال، خفیفـمتوسط، شدید و بسیار شدید تقسیم شدند. همچنین درصدNKT (CD3+ CD16/56+) خون محیطی در مواجههیافتگان و گروه کنترل به روش فلوسایتومتری بررسی شد.
نتایج: افزایش معنیدار در درصد سلولهایNKT در گروه مواجههیافته نسبتبه کنترل وجود داشت. همچنین براساس طبقهبندی GOLD، درصد سلولهای NKT یک تفاوت معنیدار در افراد مواجههیافتۀ شدید در مقایسه با گروه کنترل نشان داد.
نتیجهگیری: این سلولها توانایی مهاجرت به ریه را دارند، بنابراین این سلولهای سایتوتوکسیک که ما در خون محیطی این بیماران شناسایی کردهایم، ممکن است منعکسکنندۀ وضعیت این سلولها در ریه باشد. اگرچه مطالعات بیشتری در ریه ضروری است.

برآورد میانهدُز مؤثر و ماکزیممدُز مؤثر داروی آدرنالین بر میزان انقباض آئورت سینهای موشهای دیابتی مواجههیافته با دانۀ پسیلیوم
مقدمه و هدف: هدف اصلی توسعۀ دارو، یافتن دُز یا دامنۀ دُز دارویی است که مؤثر و ایمن باشد. یکی از اهداف مهم در مطالعات دارویی برآورد دُزهای هدف میباشد که برای این مهم معمولاً از طرح موازی استفاده میشود. هدف از این مطالعه تعیین میانهدُز و حداکثر اثر داروی آدرنالین بر میزان انقباض آئورت سینهای موشهای دیابتی که به آنها دانۀ اسفرزه داده شده بود، است.
مواد و روشها: تحقیق حاضر یک مطالعۀ تجربی با پنج دُز مختلف داروی آدرنالین بر روی چهل موش صحرایی نر سفید نژاد ویستار، در محدودۀ وزنی 8/9± 302 گرم و در محدودۀ سنی ۱۲تا۱۴ ماه میباشد. در این مطالعه، چهار مدل دُز پاسخ (خطی، ایماکس، درجهدو و ایماکس سیگموئید) برای برازش به دادهها در نظر گرفته شد. درنهایت برای انتخاب مدل از معیار آکائیک استفاده شد و تحلیل دادهها با استفاده از نرمافزار R انجام شد.
نتایج: با استفاده از معیار آکائیک (AIC) مدل ایماکس بهترین مدل شناخته شد و براساس آن میانهدُز مؤثر 8ـ10×6/6 مولار برآورد شد و حداکثر اثر با استفاده از این مدل 38/1 گرم در واحد سطح برآورد شد.
نتیجهگیری: نیمی از ماکزیمماثر میزان انقباض آئورت سینهای موشهای دیابتی که به آنها دانۀ اسفرزه داده شده، در دُز8ـ10×6/6 مولار از داروی آدرنالین رخ میدهد و باتوجهبه مقدار حداکثر اثر یعنی 38/1 گرم در واحد سطح، میتوان نتیجه گرفت که از این نقطه بهبعد هرچه میزان دُز را زیادتر کنیم، در میزان انقباض تغییری مشاهده نمیکنیم.

تأثیر 10 هفته تمرین ترکیبی (هوازی ـ مقاومتی) بر سطوح مولکول چسبان عروقی و نیمرخ لیپیدی در زنان چاق و دارای اضافهوزن
مقدمه و هدف: مولکولهای چسبان نقش مهمی در پاتوژنز آتروسکلروز دارند. هدف از پژوهش حاضر، تأثیر 10 هفته تمرین ترکیبی بر سطوح مولکولهای چسبان عروقی و نیمرخ لیپیدی در زنان چاق و دارای اضافهوزن بود.
مواد و روشها: ١٨ زن چاق و دارای اضافهوزن (میانگین سنی ٧/٧±٣٣/٣٩ سال؛ میانگین وزنی ١٩/٥±٥٧/٧٦ کیلوگرم)، در تحقیق حاضر، شرکت نمودند. آزمودنیها بهطور تصادفی به دو گروه تجربی و کنترل تقسیم شدند. آزمودنیهای گروه تجربی، تمرین ترکیبی را بهصورت تمرین هوازی (60تا75درصد ضربان قلب بیشینه) و تمرین مقاومتی (2 دوره با 1۰تا1۲ تکرار در هر جلسه و با شدت ۴0تا۶0درصد یک تکرار بیشینه و زمان استراحت ۶۰تا۹۰ ثانیه) بهمدت 10 هفته، هر هفته 3 جلسه انجام دادند. در طی پژوهش، گروه کنترل هیچ مداخلهای دریافت نکردند. در آغاز و ٤٨ ساعت پس از پایان تمرین، خونگیری بهمنظور اندازهگیری سطوح مولکول چسبان عروقی (VCAM-1) و لیپیدها انجام گرفت. دادهها با استفاده از آزمون تی مستقل در سطح ٠٥/٠p< با استفاده از نرمافزار 20SPSS تحلیل شد.
نتایج: نتایج نشان داد که بین دو گروه کنترل و تجربی در تغییرات ترکیبات بدن، چربیهای خون و سطوح مولکول چسبان عروقی تفاوت معناداری مشاهده نشد (005/0p>).
نتیجهگیری: بهنظر میرسد 10 هفته تمرین ترکیبی بهمنظور کاهش سطوح VCAM-1 و چربیهای خون کافی نباشد و نیاز به تحقیقات بیشتری در این زمینه میباشد.

بررسی اثر عصارۀ پلاکتی مشتق از خون بندناف بر تکثیر سلولهای تکهستهای خون بندناف در آزمایشگاه
مقدمه و هدف: علیرغم نتایج رضایتبخش پیوند خون بندناف در درمان بعضی بیماریهای خونی، یکی از چالشها در این زمینه، حل مشکل تعداد کم این سلولها میباشد. سالهای اخیر عصارۀ پلاکتی بهلحاظ غنیبودن از فاکتورهای رشد بهعنوان مکمل کشت سلول مزانشیم در سلول درمانی معرفی شده است. هدف این مطالعه، بررسی اثر عصارۀ پلاکتی مشتق از خون بندناف بر تکثیر، تمایز و درصد زندهمانی سلولهای تکهستهای خون بندناف در شرایط آزمایشگاهی است.
مواد و روشها: کنسانترۀ پلاکتی در غلظت plt/ml109×2 از پلاسمای خون بندناف تهیه شد. تعداد 106 سلول تکهستهای خون بندناف در حضور سایتوکاینهای Flt3L، SCFوTPO حاوی غلظتهای متفاوت (10%-0) از عصارۀ پلاکتی مشتق از خون بندناف کشت شدند. درصد زندهمانی این سلولها با استفاده از رنگآمیزی تریپان بلو، سنجش تمایز سلولهای تکهستهای آزمایش Colony Assay، اندازهگیری درصد سلولهای بنیادی خونساز با شاخص سطحی (CD133+ CD34+ CD38-) از طریق فلوسایتومتری و بیان کمی ژن CD34 با روشQRT- PCR مورد ارزیابی قرار گرفت.
نتایج: غلظت 5٪ عصارۀ پلاکتی مشتق از خون بندناف همراه با سایتوکاینها سبب تکثیر 12برابری (P<0.0001) سلولهای تکهستهای خون بندناف، 6/1 برابر کلونی BFU-E، 1/2 CFU-GM، 7/1 CFU-MIX و 3/3 برابر سلولهایCD133+ CD34+ CD38- میشود. ژن CD34 نیز در گروه ۵٪ LP -UCB افزایش معنیدار نشان داد.
نتیجهگیری: عصارۀ پلاکتی خون بندناف را میتوان بهعنوان مکمل در کشت سلولهای تکهستهای خون بندناف و سلولهای بنیادی خونساز بهمنظور افزایش تکثیر و حفظ خاصیت بنیادی آنها استفاده کرد.

بررسی ارتباط بین شدت بیثباتی خودگزارشی با تعادل و سطح سلامت در افراد با بیثباتی عملکردی مچ پا
مقدمه و هدف: بیثباتی مزمن عملکردی مچ پا که با حس بیثباتی و تکرار پیچخوردگی مشخص میشود، با اختلال تعادل و محدودیت در فعالیتهای روزمره و ورزشی افراد، سطح سلامت آنها را دستخوش تغییر میکند. اطلاعات کمی در مورد رابطۀ بین احساس بیثباتی گزارششده توسط افراد، که شکایت اصلی بیماران است، با وضعیت تعادل و سطح سلامت آنها موجود میباشد.
مواد و روشها: 23 فرد مبتلا به بیثباتی مزمن یکطرفۀ مچ پا (63/4±31/29 سال) و 23 فرد سالم (21/5±52/30 سال) در این مطالعه شرکت کردند. جهت سنجش تعادل از مجموعه آزمونهای Balance Error Scoring System (BESS) استفاده شد. همچنین افراد دو پرسشنامۀ SF-36 و شاخص بیثباتی مچ پا را تکمیل نمودند. برای مقایسه بین گروهها از آزمون تی مستقل و من ویتنی و برای ارتباطسنجی بین متغیرها از آزمون همبستگی اسپیرمن استفاده شد.
نتایج: نمرۀ BESS در گروه افراد با بیثباتی عملکردی مچ پا بهطور معنیداری بیشتر از گروه کنترل بود (P<0.05). آزمون همبستگی اسپیرمن نشان داد که رابطۀ معنیدار بین شاخص بیثباتی مچ پا با تمام بخشهای پرسشنامه SF-36 بهجز انرژی/خستگی و عملکرد اجتماعی وجود دارد (r= 0.43 to 0.85, P>0.05). هیچ رابطۀ معنیداری بین آزمونهای تعادلی با شاخص بیثباتی مچ پا یافت نشد.
نتیجهگیری: ارتباط قوی شاخص بیثباتی با بخش جسمانی پرسشنامه SF-36نشان میدهد میزان بیثباتی که توسط بیمار گزارش میشود، تأثیر جدی بر سلامت جسمانی او میگذارد. اما گزارش بیمار با نتایج آزمونهای تعادلی ارتباط معنیدار ندارد.
