Capsule production as a means of resistance can prevent host immune system responses. This study investigated the effect of a PLGA nanoparticles vaccine containing capsule antigen.Materials and Methods: Vaccination of 20 mice with an age of 3-5 weeks (with a weight range of 22±18 g) was performed in four groups of capsules, PLGA, PLGA-capsule combination (PLGA+CPS) and control (PBS). In this study, the fabrication of nanoparticles was investigated by zetasizer and FITR tests. Microbial loading was also performed on the bladder.One-way analysis of variance and Tukey post hoc test were used for statistical analysis .Results: The results of confirmation tests of zetaiser showed that the nanoparticle size and the size of PLGA nanoparticles containing capsule antigen molecule were 178.7 and 159.4 nm, respectively.The result of FTIR and the shapes of the corresponding peaks confirmed the presence of antigen functional groups in the nanoparticle structure and the formation of ester bonds.The resut of FTIR test also indicates the success of PLGA nanoparticles containing capsule antigen , the results of microbial challenge showed that in the control and PLGA groups, due to the fact that PLGA nanoparticles vaccine contain capsule antigen were not used, a significant increase was observed in terms of the number of colonies compared to other groups  )p<0/05( .Conclusion: PLGA nanoparticles containing capsule antigen molecules have a better ability than pure capsules and can significantly reduce tissue damage.}, keywords = {Klebsiella pneumoniae,PLGA nanoparticles,vaccine containing capsule antigen,Capsule antigen,PLGA,Microbial}, title_fa = {اثر ایمنی زایی نانوپارتیکل PLGA حاوی آنتی ژن کپسول کلبسیلا پنومونیه K2O1 علیه عفونت ادراری در موش BALB/C}, abstract_fa = {مقدمه و هدف: کلبسیلا میتواند باعث عفونت های ادراری شود که درمان آن با مقاومت دارویی مواجه شده است. کپسول به عنوان یکی از راههای مقاومت میتواند از پاسخ های سیستم ایمنی میزبان جلوگیری کند. این مطالعه، اثر واکسن نانوپارتیکل  PLGA حاوی آنتی ژن  کپسول را مورد بررسی قرار داده است.مواد و روش ها: واکسیناسیون 20 سر موش 3-5 هفته ای (محدوده وزنی 18 ±22 گرم)، در چهارگروه کپسول،PLGA ، PLGA حاوی کپسول و کنترل انجام شد. در این مطالعه، ساخت نانوپارتیکل با آزمون های زتاسایزر و FITR بررسی گردید. چلنج بار میکروبی نیز بر روی مثانه انجام شد. آزمون آنالیز واریانس یک طرفه و آزمون تعقیبی توکی برای تحلیل آماری استفاده گردید.نتایج: نتایج آزمون های زتاسایزر نشان داد که اندازه نانوپارتیکل و اندازه PLGA حاوی انتی ژن کپسول به ترتیب 178.7 و 159.4 نانومتر بدست آمد .نتایج FTIR وشکل پیک های مربوطه ،حضور گزوه های عاملی آنتی ژن در ساختار نانوپارتیکل و تشکیل پیوند استری را تایید کردندو نتیجه FITR نشان دهنده موفقیت آمیز بودن PLGA حاوی کپسول می باشد. نتایج چلنج میکروبی نشان داد که در گروه های کنترل و  PLGAبا توجه به اینکه از واکسن های PLGA حاوی کپسول استفاده نشده بود افزایش معنی داری با توجه به تعداد کلنی ها در مقایسه با سایر گروه ها مشاهده شد (p<0/05).نتیجه گیری: مولکول های PLGA حاوی آنتی ژن کپسول توانایی بهتری نسبت به کپسول خالص داشته و می توانند به طور معناداری کاهش تخریب بافتی را بدنبال داشته باشند.}, keywords_fa = {کلبسیلا پنومونیه,واکسن PLGA حاوی آنتی ژن کپسول,آنتی ژن کپسول,PLGA,چلنج میکروبی}, url = {https://daneshvarmed.shahed.ac.ir/article_3517.html}, eprint = {https://daneshvarmed.shahed.ac.ir/article_3517_654e9693dc98966c3637940a453ea139.pdf} }
