Recently, identification of menstrual blood-derived stem cells (MenSCs) as a unique source of stem cell with some characteristics as well as ease of access, high proliferative ability and renewability has created enormous promise for cell therapy. Materials and Methods: In this study, differentiation ability of MenSCs into cardiomyocytes has been investigated. After MenSCs immunophenotyping, their differentiation into cardiomyocyte was investigated in the presence of 5-azacytidine and basic-fibroblast growth factor. Then, expression of the putative myogenic cells at mRNA and protein levels was determined by immunofluorescent staining and real-time quantitative PCR. Results: Based on flow cytometric analysis, the isolated MenSCs typically expressed mesenchymal stem cell markers like CD105, CD73, CD44 and CD166 in parallel to OCT-4 as an embryonic marker. The differentiated MenSCs expressed cardiomyocyte markers at mRNA/protein level. The myogenic cells differentiated from MenSCs were positive for Connexin-43 and troponin T2 (TNNT2) protein. The mRNAs of Connexin-43, Connexin-40, Alpha actinin, Tropomyosin1 and TNNT2 were highly expressed in the differentiated myogenic cells. Conclusion: Based on our data, MenSCs are a unique cell population with differentiation ability into cells with characteristics commonly attributed to cardiomyocytes. }, keywords = {Menstrual blood,Stem cell,Cardiomyocyte,Differentiation}, title_fa = {ارزیابی توان تمایز سلول های بنیادی مشتق از خون قاعدگی به سلولهای قلبی در شرایط آزمایشگاهی}, abstract_fa = {مقدمه و هدف: در دهه‌های اخیر، سلول‌های بنیادی به‌عنوان یک روش درمانی جدید برای بیماران مبتلا به اختلالات قلبی معرفی شده است. به‌تازگی شناسایی سلول‌های بنیادی مشتق از خون قاعدگی به‌عنوان یک منبع منحصربه‌فرد از سلول‌های بنیادی با برخی خصوصیاتی مثل سهولت دسترسی، توانایی تکثیر و خودتجدیدی بالا، امید فراوانی را برای سلول‌درمانی ایجاد کرده است. در این مطالعه، توانایی تمایز این سلول‌ها به کاردیومیوسیت بررسی شده است. مواد و روش‌ها: بعد از بررسی مارکرهای سطحی این سلول‌ها تمایز آن‌ها به سلول‌های شبه کاردیومیوسیت در حضور 5- آزاسیتیدین و فاکتور رشد فیبروبلاستی بررسی شد. سپس بیان سلول‌های تمایزی در سطح پروتئین و mRNA به‌وسیلۀ رنگ‌آمیزی ایمونوفلورسنت و real-time quantitative PCR بررسی گردید. نتایج: براساس آنالیز فلوسایتومتری ، این سلول‌ها به‌طور معمول، مارکرهای سلول‌های بنیادی مزانشیمی شبیه CD105، CD73، CD44و CD166 همچنین OCT-4 به‌عنوان یک مارکر جنینی را بیان کردند. سلول‌های تمایزداده‌شده، مارکرهای سلول‌های کاردیومیوسیتی را در سطح mRNA و پروتئین بیان کردند. سلول‌های کاردیومیوسیت تمایزیافته از این سلول‌ها برای پروتئین Connexin-43 و troponin T2 (TNNT2) مثبت بودند. همچنین mRNA ، Connexin-43،Connexin-40، Alpha actinin، Tropomyosin1 و TNNT2 در سطح بالایی در سلول‌های تمایزی بیان گردید. نتیجه‌گیری: براساس داده‌های ما سلول‌های بنیادی مشتق از خون قاعدگی، جمعیت سلولی منحصربه‌فردی هستند که توانایی تمایز به سلول‌هایی با ویژگی‌هایی که به‌طور معمول، به سلول‌های کاردیومیوسیت نسبت داده می‌شود دارند. }, keywords_fa = {خون قاعدگی,سلول بنیادی,کاردیومیوسیت,تمایز}, url = {https://daneshvarmed.shahed.ac.ir/article_1728.html}, eprint = {https://daneshvarmed.shahed.ac.ir/article_1728_f36074c52980f2170c67ec3fdad4a10d.pdf} }
