The Leishmania expression system represents the combination of easy handling known from bacterial expression systems with the potential of a eukaryotic protein expression, folding and modification system. The trypanosomatid protozoan host Leishmania tarentolae was isolated from lizard and is not pathogenic to mammals.    Materials and Methods: In this study, two constructs were developed for expression of recombinant human interferon-gamma (hrIFNg) in L. tarentolae. Each one of constructs carries an antibiotic resistant gene such as Hygromycin and Nourseothericin. For high level expression of the human interferon-gamma, developed DNA cassette that contains interferon-gamma (IFNg) cDNA was designed to integrate into a genomic small subunit rRNA locus of L. tarentolae by homologous recombination.   Results: The integration of the expression cassette into the ssu locus was confirmed by diagnostic PCR of the genomic DNA of transgenic strain.    Conclusion: Although it has been shown that E. coli might be considered as a suitable host with a relatively high level of expression and lack of posttranslational modifications and the need for refolding stages are still big challenges. In contrast, L. tarentolae is eukaryotic gene expression machinery, which includes full glycosylation and disulfide bond formation, and thus represents a potential advantage over other expression systems. These facts confirm that the gene expression system using Leishmania parasites combines many of the advantages of both prokaryotic and eukaryotic expression systems.  }, keywords = {hrIFNg,ssu locus,L. tarentolae}, title_fa = {تهیه سازه‌های مناسب به‌منظور بیان ژن اینترفرون گامای انسانی در Leishmania tarentolae}, abstract_fa = { مقدمه و هدف: افزایش کاربردهای درمانی برای اینترفرون گامای انسانی نوترکیب (hrIFNg) که یک سایتوکین پیش­التهابی ضدویروسی است، موجب گسترش روش­های بهینه­سازی تولید آن شده­است؛ در این مطالعه، تولید این گلیکوپروتئین در سیستم یوکاریوتی تک­سلولی Leishmania tarentolae که از مارمولک tarentolae annularis به­دست­می­آید، برای بیان ژن هترولوگ مورد بررسی قرارگرفته­است.    مواد و روش­ها: به­منظور تکثیر cDNA ژن IFNg از روش RT-PCR روی سلول­های تک­هسته­ای خون محیطی تحریک­شده با فیتوهماگلوتینین یک فرد ایرانی سالم استفاده­شد و به­منظور بیان پروتئین hrIFNg، cDNA تکثیرشده در دو کاست بیانی کلون شد. به نحوی که هر کاست بیانی دارای یک کپی از ژن hIFNg بود؛ این کاست­های بیانی با روش الکتروپوریشن در جایگاه ژن RNA ریبوزومی (ssu) L. tarentolae وارد شدند. پس از ترانسفورم، کلون­های ترانسفورم­شده در حضور آنتی­بیوتیک­های اختصاصی انتخاب­شدند.    نتایج: سازه­های مورد نیاز بیان اینترفرون گامای انسانی در سلول­های L. tarentolae تهیه و به داخل سلول انگل وارد شد و کارایی سازه­ها، با تکنیک وسترن بلات تأییدشد.   نتیجه­گیری: تاکنون انواع مختلفی از سلو­ل­های پروکاریوتی و یوکاریوتی به­عنوان سیستم­های بیانی در تولید پروتئین­های نوترکیب استفاده­شده­اند ولی به دلیل ویژگی­هایی، اغلب در بیان این گلیکوپروتیین ناموفق بوده­اند. سیستم به­کار­رفته در این مطالعه از خصوصیاتی ویژه­ ازقبیل سرعت رشد بالا، شرایط کشت ارزان و گلیکوزیلاسیون مناسب گلیکوپروتیین­ها بهره­مند است که می­تواند برای تولید پروتئین نوترکیب به­کاررود. }, keywords_fa = {اینترفرون گامای انسانی نوترکیب,جایگاه ریبوزومی (ssu),لیشمانیا تارنتولی}, url = {https://daneshvarmed.shahed.ac.ir/article_1526.html}, eprint = {https://daneshvarmed.shahed.ac.ir/article_1526_2550b0047a207f761c2123ba5e2337db.pdf} }
