In this new kind of chimeric T cell receptor, nanobodies are replaced as variable fragment of T cell receptor. Nanobodies (VHH) are the smallest fragments of antibodies that have great homology to human VH and low immunogenic potential. VHH-hing-CD28-CD3و construct was made in our laboratory. Tumor cells rarely provide costimulatory signals and hence CAR receptors that transmit just a CD3و signal can only initiate target cell killing and interferon γ release and fail to induce full activation. Although incorporation of a CD28 component results in IL-2 (Interlukin 2) release and limited proliferation, T cell activation remains incomplete. To optimize CAR signaling, tripartite endodomains were constructed which consist of CD28-OX40-CD3ζ. During T cell activation, OX40 transmits a potent and prolonged T cell activation signal and it is crucial for maintaining an immunological response. Materials and Methods: We designed overlapping primers that contain 60% of OX40 sequence, then, CD28-Ox40-CD3ζ region was synthesized by SOE-PCR, consequently, it was replaced with CD28-CD3ζ region in VHH-Hing-CD28-CD3ζ construct. Then, this construct was transfected in Jurkat T cells, so IL-2 secretion and T cells proliferation were increased. Results: CD28-OX40-CD3ζ tripartite cytoplasmic domain provided a full complement of activation, proliferation, and survival signals for enhanced anti-tumor activity. Conclusion: With IL-2 and semi-quantitative RT PCR, it is concluded that mRNA expression of pCDNA-1-hinge-CAR-OX40 and pCDNA-2-hinge-CAR-OX40 constructs are the same but the function of pCDNA-2-hinge-CAR-OX40 construct is more than the others.}, keywords = {chimeric T cell receptor,Nanobody,OX40,T lymphocyte}, title_fa = {ساخت گیرنده کایمریک لنفوسیت T دارای کمک محرک OX40 علیه سلول‌های سرطان سینه}, abstract_fa = {مقدمه و هدف: گیرنده­های کایمریک سلول­های T روش خوبی برای ایمنوتراپی سرطان فراهم‌کرده‌است. در این گیرنده بجای قطعه متغیر گیرنده سلول T از نانوبادی استفاده‌شده‌است. نانوبادی یا VHH کوچکترین قطعه آنتی‌بادی زنجیره سنگین شتری است که قابلیت اتصال به آنتی­ژن دارد و نیز همولوژی بسیاری با VH انسانی داشته، از قدرت ایمنی زایی اندکی بهره‌مند است. سازه رسپتور کایمریکی که در آزمایشگاه ما ساخته‌شده دارای VHH-hing-CD28-CD3ζ است. اگرچه گیرنده­های کایمریکی که از طریق CD3ζ فعال می­شوند اینترفرون گاما آزاد می­کنند، اما به علت نبود سیگنال­های کمک تحریکی  نمی­توانند به طور کامل فعال شوند. لذا وجود CD28 باعث رهاسازی اینترلوکین 2 شده و تکثیر تومور را محدود می­کند؛ اما باعث فعال شدن کامل T سل نمی­شود. به‌منظور بهبود عملکرد گیرنده­های کایمریک، اندودامین­های سه بخشی ساخته‌شده‌است که شامل CD28-OX40-CD3ζ است. به دنبال تحریک سلول­های T کمک محرک OX40 سیگنال­های قوی فعا‌ل‌سازی و طولانی‌مدت برای این سلول­ها ایجادمی­کند که برای حفظ پاسخ ایمنی بسیار مهم است.مواد و روش­ها: در این تحقیق پرایمرهای هم­پوشان که 60% از توالی OX40 را در برداشت سنتز و قطعه CD28-Ox40-CD3ζ با روش SOE PCR ایجاد شد و سپس این توالی با قطعه  CD28-CD3ζ در سازه  VHH-hing-CD28-CD3ζ جایگزین شد. بعد از انتقال این سازه به درون سلول­های Jurkat) T) بیان گیرنده در سطح mRNA و عملکرد آن با روش­های semi-quantitative RT PCR و کیت الایزا سنجش IL-2 ارزیابی‌شد.نتایج و نتیجه­گیری:  نتایج حاکی از آن است که عملکرد و بیان سازه دارای OX40 بیشترشده‌است. با توجه به نتایج ارزیابی IL-2 و semi-quantitative RT PCR می­توان چنین نتیجه‌گیری‌کرد که میزان بیان سازه‌های pCDNA-1 hinge CAR-OX40 و pCDNA-2 hinge CAR-OX40 برابرند اما عملکرد سازه pCDNA-2 hinge CAR-OX40 بیشتر است. }, keywords_fa = {گیرنده سلول T کایمریک,نانوبادی,OX40,Tلنفوسیت}, url = {https://daneshvarmed.shahed.ac.ir/article_1479.html}, eprint = {https://daneshvarmed.shahed.ac.ir/article_1479_bbf4b9d152feab0487d2aef03be5f2aa.pdf} }
